Two-Window Approach to Monitor and Assess Cellular and Humoral Immune Responses in Poultry

As previously reported, inflammatory activity initiated by intradermal injection of multiple growing feather (GF)-pulps a chicken with lipopolysaccharide, and the subsequent periodic sampling GFs blood, enables longitudinal evaluation in vivo tissue- systemic-inflammatory activities ex laboratory analyses. To demonstrate suitability this two-window approach to monitor assess vaccine responses, two groups chickens were immunized intramuscular mouse IgG (mIgG), mIgG alum adjuvant (Alum&mIgG), or PBS-vehicle (Group I II at 7- 11-weeks, respectively). Plasma levels mIgG-specific antibodies monitored ELISA for 28 days post-primary- secondary-immunizations. examine cellular 20 GF-pulps per bird injected on Day-10 Day-5 -secondary-immunization, respectively. Two collected before- various times (0.25 7 days) post-injection leukocyte population- cytokine mRNA expression-analyses. The observed primary- secondary-antibody response profiles as expected T-dependent antigen. Leukocyte- cytokine-profiles established revealed temporal, qualitative, quantitative differences local naïve, primary, secondary leukocyte-effector responses This study demonstrates unique opportunity avian model both cell- antibody-mediated immune using minimally invasive techniques.
